Food Nutr Res:伊贝母总生物碱提取物对香烟烟雾致Beas-2B细胞损伤模型的保护作用及转录组学分析

该研究探讨伊贝母(BFP)总生物碱提取物(BFP-TA)对香烟烟雾提取物(CSE)诱导的Beas-2B细胞损伤的作用及机制。

背景:慢性阻塞性肺病(COPD)是一种慢性进行性气道和肺部疾病,患病率和死亡率都很高,而且患病率随着年龄的增长而增加。据世界卫生组织预测,到2030年,慢性阻塞性肺病预计将成为全球第三大死亡原因。目前的药物治疗对COPD的进展或死亡率影响有限,而且几乎没有证据表明任何一种药物具有逆转气道重塑和气流阻塞的潜力。因此,迫切需要开发安全有效的COPD治疗新药。

伊贝母(BFP)是一种长期用于治疗肺部疾病的中药,具有润肺、化痰、止咳、抗炎、抗氧化等功效,但其有效成分和作用机制尚不清楚。

目的:探讨BFP总生物碱提取物(BFP-TA)香烟烟雾提取物(CSE)诱导的Beas-2B细胞损伤的作用及机制。

方法:用2% CSE诱导Beas-2B细胞损伤模型,按照T-AOC检测试剂盒、SOD检测试剂盒和MDA检测试剂盒的说明检测BFP-TA对细胞总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平的影响,荧光显微镜下检测ROS的产生。流式细胞术检测BFP-TA对Beas-2B细胞凋亡的影响,western blot检测BFP-TA对相关蛋白表达的影响。

随后,通过转录组测序研究了BFP-TA对CSE诱导的Beas-2B细胞中差异表达基因(DEGs)的影响,并通过定量实时聚合酶链反应(qPCR)验证了DEGs的表达。

结果:BFP-TA通过提高T-AOC和SOD水平,抑制ROS和MDA水平,减轻CSE诱导的Beas-2B细胞氧化损伤,其机制可能与SIRT1/Nrf2/Keap1信号通路有关。此外,BFP-TA可以通过抑制Bax、MST1和FOXO3a蛋白的表达来抑制CSE诱导的细胞凋亡,并通过抑制MAPK信号通路的激活来发挥抗炎作用

随后,转录组分析和qPCR验证表明,BFP-TA通过调节PPAR和PI3K-Akt信号通路中DEGs的表达,减轻炎症、氧化应激、细胞凋亡和脂质代谢紊乱,从而对CSE诱导的Beas-2B细胞损伤起到保护作用

结论:该研究首次证实BFP-TA可通过抗炎、抗氧化、抗凋亡、调节脂质代谢紊乱等作用对CSE诱导的Beas-2B细胞损伤起到保护作用。
