自然杀伤细胞（NK）是一种淋巴样细胞群，在调节天然免疫反应中发挥着重要作用。在小鼠的研究中表明NK细胞在肾病模型中发挥着病理作用。近日，肾脏病领域权威杂志kidney international上发表了一篇研究文章，在这项研究中，研究人员旨在归纳肾小管间质纤维化患者NK细胞亚群的特点，以及明确慢性肾脏疾病的病理标志。

研究人员通过多色流式细胞术发现肾小管间质纤维化患者肾活检中NK细胞（CD3-CD56+）总数显著增加，相比于无纤维化病变肾活检和健康肾组织。在亚组水平，CD56dim NK细胞亚群，特别是CD56bright NK细胞亚群在肾纤维化组织中明显增加。然而，只有CD56bright NK细胞与肾功能的损失显著相关。纤维化的肾活检组织中CD56bright NK细胞组织保持和活化分子CD69明显增加，相比于非纤维化的肾组织，提示为致病表型。

进一步的流式细胞表型显示在CD56bright NK细胞中激活受体CD335（NKp46）和分化标记CD117（c-kit）的选择性共表达。纤维化肾组织的多色免疫荧光染色定位了NK细胞积聚位于肾小管间质，CD56bright NK细胞（NKp46+ CD117+）被确定为NK细胞间室内促炎细胞因子干扰素-γ的来源。

因此，激活干扰素-γ产生的CD56bright NK细胞能够在纤维化过程和进展为慢性肾脏病中发挥关键作用。
