最近,来自美国西南医学中心的研究人员成功解析了一种在多种癌症特别是血液癌症发展中发挥重要作用的酶复合体的原子结构。相关研究结果发表在国际学术期刊science上。
 
这一被成功解析的酶复合体叫做polycomb repressive complex2(PRC2),是参与人类发育的一种关键调控因子,它能够通过改变染色质结构调节基因表达。作为一种酶复合体,PRC2能够对组成染色质的组蛋白进行修饰,导致染色质结构发生关键性改变,从而沉默特定基因的表达。之前研究已经发现PRC2的异常表达与淋巴瘤,白血病,脑瘤以及其他疾病的发生有关。
 
研究人员表示,PRC2表达过高或过低会导致基因发生相应的沉默或激活,这对于细胞来说不是一件好事,这项研究结果揭示了正常水平的PRC2如何在细胞内保持酶活性以及如何受到调控。该研究人员还指出,他们发现了PRC2发挥作用的不同方式,这对于我们了解相关疾病的化学基础以及开发疾病治疗药物都非常具有帮助。目前一些抑制PRC2酶活性并用于治疗特定类型淋巴瘤的小分子药物已经得到开发。
 
这项研究为进一步研究PRC2在正常细胞和疾病细胞中的功能和调节奠定了基础,研究人员表示,他们正在对PRC2 复合体与染色质相互作用过程中,细胞对PRC2的调控过程以及该复合体如何与其他细胞蛋白相互作用影响染色质结构,开启和关闭特定基因表达进行深入研究。
 
这项工作可能会为接下来进行PRC2功能及其在人类疾病中的作用研究提供更加全面深入的理解。
