抗白血病化合物MI-3454可诱导小鼠模型完全缓解

美国密歇根大学开发的一种抗癌化合物在小鼠模型中显示出对白血病的高度活性,该化合物被称为MI-3454,可抑制MeninMLL1蛋白之间的蛋白质相互作用。将具有高表达MLL1基因的患者来源的肿瘤细胞移植进小鼠体内,MI-3454诱导了小鼠体内肿瘤的完全缓解。成人急性白血病患者中有5%至10%具有MLL1基因重排,婴儿急性淋巴细胞白血病中有80%具有MLL1基因重排。

该化合物还导致具有NPM1基因突变的白血病模型中的淋巴母细胞大量减少,NPM1基因突变存在于大约30%的急性髓细胞性白血病病例中。密歇根州立大学的病理学副教授Jolanta Grembecka博士说:只有约三分之一的MLL1基因重排患者能够存活五年,因此迫切需要新的治疗方法
