虽然许多神经退行性疾病相关蛋白可以在线粒体中发现,但目前仍不清楚线粒体功能障碍和蛋白质聚集是如何相关的。

在分裂的酵母细胞中,应激条件下或衰老期间形成的蛋白质聚集体优先被母细胞保留,部分通过栓系到线粒体中,而解聚体Hsp104有助于解离聚集体,从而能够重新折叠或降解错误折叠的蛋白质。

近期,在此项发表在权威杂志Nature上的研究显示,在酵母中,易于聚集的胞质蛋白质被导入线粒体进行降解。在热休克下形成的蛋白质聚集体包含胞质和线粒体蛋白质并与线粒体复合物相互作用。

许多易于聚集的蛋白质在热休克后进入线粒体间隙空间和基质,一些甚至没有经过应激。及时溶解胞质聚集物需要线粒体和蛋白酶。阻断线粒体输入而不是蛋白酶体活性导致聚集蛋白质降解的显着延迟。

胞质Hsp70s缺陷导致错误折叠的蛋白质进入线粒体的增加以及线粒体应激的升高。

研究者们称这种线粒体介导的蛋白质稳态机制为MAGIC(线粒体作为细胞质中的守护者),并且提供了它可能存在于人类细胞中的证据。
