癌症的扩散需要脂肪做燃料

过去,人们一直认为糖是癌细胞扩散的主要能量来源,但今年早些时候的一项研究表明,我们在癌细胞转移上的研究一直是错误的,可能是脂肪推动的癌细胞的扩散。


现在这项新研究给这个假设增加了有力的证据。


研究人员发现,小鼠体内负责口腔癌扩散的细胞实际上的能量来源是脂肪酸,包括棕榈酸——一种常见的食品添加剂,是棕榈油的主要组成成分。


他们是在注意到许多转移细胞中一种叫做CD36的受体蛋白表达水平较高,这种蛋白帮助细胞吸收油脂。


CD36的高表达也与癌症患者较差的治疗效果有关,所以研究团队决定看看受体被阻止时会发生什么。


难以置信的是,当癌细胞中CD36的表达受阻时,能够完全阻止人类的癌症在小鼠体内扩散,但它却没有阻止肿瘤的形成。


研究的领导者Salvador Aznar Benitah对Research Gate说道:“我们假设负责扩散的细胞依赖某种特定的脂肪酸,没有这些脂肪酸它们无法工作。但我们还不知道为什么阻止了CD36就会产生此后果其中的机制。”


研究团队还有许多工作要做,目前的结果表明当癌症扩散后这种方法也能起到一定的作用。


阻止小鼠体内CD36的表达使肿瘤的扩散减少了15%,剩下的肿瘤也减小了80%。


研究还发现,相比于正常饮食的小鼠高脂肪的饮食让它们淋巴结和肺部的肿瘤变得更大,这是一个肿瘤扩散的信号。


需要澄清的是,此研究只针对小鼠体内的人类癌症细胞,并不保证同样的方法对人类也有效。


目前这个阶段,没有人会建议减少食物中的脂肪摄入以避免癌症的扩散,特别是对于癌症病人来说需要高能量的饮食来保持健康。


研究团队目前正在努力开发适用于人类的对抗CD36的抗体,希望未来5年内能够进行临床试验。


Benitah说:“这是令人兴奋的并且十分重要的第一步。我们已经识别出负责转移癌症的细胞,接下来将对它们的行为进行更加详细的研究。”


他补充到:“它打开了一种抗转移的新疗法。”


我们期待这项研究的最终结果,因为对于治疗癌症我们已经有了许多革命性的新疗法,但阻止它的扩散还是史无前例的第一次。

科学家探明非酒精性脂肪肝发病机理

近日,合肥工业大学科研团队在细胞与分子水平上取得一项研究成果,探明了非酒精性脂肪肝发病机理,发现了新的调控肝脏脂肪化基因。该成果刊登在《肝脏学》上。

非酒精性脂肪肝病因复杂,对应的治疗手段非常有限。合肥工业大学***特聘教授韩际宏课题组,与美国威斯康辛医学院开展合作研究,发现了新的调控肝脏脂肪化的基因Nogo-B蛋白受体,有望为非酒精性脂肪肝的治疗提供新的药物靶点。

Nogo-B蛋白受体又称NgBR。该项研究发现,肝脏中NgBR蛋白水平降低与脂肪肝发病有直接关系。在相关实验中,敲除NgBR基因的小鼠肝脏中积累了大量甘油三酯与自由脂肪酸。研究证实,这种脂肪的积累是由于NgBR水平下降导致调控肝脏脂肪酸合成的中枢分子肝X受体alpha的核转移与激活,同时与具有增强体内能量代谢传感器功能的腺苷酸活化蛋白激酶alpha失活密切相关。
