成牙本质细胞贯穿一生分泌形成牙本质,并由此应对外伤。这篇研究的目的是为了评估内源性Wnt信号在调控牙本质累积率中的作用。

研究采用组织学、组织化学、活体染色和组织形态学鉴定来定量作为增龄功能之一的牙本质累积率。采用2株Wnt报告基因小鼠来鉴定和追踪作为增龄功能之一的Wnt反应阳性成牙本质细胞的分布和数量。为了显示牙本质分泌和Wnt信号之间的因果关系,在1株Wnt信号显著上调的小鼠中检测牙本质累积情况。

结果显示,牙本质沉积贯穿一生,但沉积率随着年龄增加而减慢。这种牙本质分泌的减少与内源性Wnt信号减少相关联。在基因修饰过的小鼠中,异常上调Wnt信号导致前成牙本质细胞分泌修复性牙本质或骨性牙本质累积的增加,进而替代管状牙本质。

结论:Wnt信号通过成牙本质细胞调控牙本质的分泌,并且上调Wnt信号直接促使修复性或骨性牙本质的形成。这些临床前数据对于研发生物类盖髓材料具有一定的治疗意义。
