Int J Mol Sci：新型水凝胶可有效促进骨再生

对于骨再生，使用生物相容性热凝胶水凝胶，透明质酸-g-壳聚糖-g-聚（N-异丙基丙烯酰胺）（HA-CPN）作为三维有机凝胶基质，用于捕获兔脂肪来源的干细胞（rASCs）。将双相磷酸钙（BCP）陶瓷微粒作为矿化骨基质嵌入凝胶基质中，进一步使用具有骨诱导性能的富含血小板的血浆（PRP）强化。比较HA-CPN和HA-CPN/PRP/BCP中rASCs的体外培养细胞增殖和成骨分化。结果显示，HA-CPN/

将双相磷酸钙（BCP）陶瓷微粒作为矿化骨基质嵌入凝胶基质中，进一步使用具有骨诱导性能的富含血小板的血浆（PRP）强化。比较HA-CPN和HA-CPN/PRP/BCP中rASCs的体外培养细胞增殖和成骨分化。

结果显示，HA-CPN/PRP/BCP是用于rASCs成骨的更好的可注射细胞载体，可增加细胞的增殖率和碱性磷酸酶活性，增强钙沉积和细胞外基质的矿化，以及上调成骨基因标记物的表达。在兔临界尺寸大小的颅骨缺损中植入HA-CPN/PRP/BCP/rASCs构建体，通过计算机断层扫描，组织学和免疫组织化学分析成功地证实了缺损部位的新骨形成。

总之，通过将PRP和BCP结合为HA-CPN的骨诱导和骨传导因子，我们成功地证明了热凝胶复合水凝胶支架可以促进rASCs的骨生成，可用于骨组织工程。
