iMatrix-511是一个全新的来源于laminin-511的整联蛋白结合片段。之前的研究显示，它作为培养基底物对无添加培养基具有很大的优越性，并且可以维持干细胞的多能性。然而，它在牙科领域的研究还所知甚少。这篇研究的目的是为了阐明iMatrix-511在体外的作用，并与玻连蛋白(VN)相比较。

iMatrix-511是一个全新的来源于laminin-511的整联蛋白结合片段。之前的研究显示，它作为培养基底物对无添加培养基具有很大的优越性，并且可以维持干细胞的多能性。然而，它在牙科领域的研究还所知甚少。这篇研究的目的是为了阐明iMatrix-511在体外的作用，并与玻连蛋白(VN)相比较。

研究使用MDPC-23细胞进行生物化学的检测。采用细胞计数kit-8法确定2种蛋白的最佳涂布浓度。为了评估2种蛋白对增殖的影响，研究将MDPC-23细胞直接接种于iMatrix-511 或改良VN聚苯乙烯上并通过CCK-8进行检测。鉴定接种于iMatrix-511和VN细胞的表型。确定表型特征的方法包括实时聚合酶链式反应和茜素红染色。

结果显示，iMatrix-511和VN的最佳涂布浓度分别为1 μg/cm2和0.25 μg/cm2。在培养的1，2和4天时，培养在iMatrix-511细胞的增殖活性要明显高于对照和VN组。细胞形态学的观察显示，MDPC-23细胞首先粘附于iMatrix-511上，并且早在接种1小时就开始扩增。培养于iMatrix-511上MDPC-23的牙本质基质蛋白1和牙本质涎磷蛋白信使RNA的表达显著增加，表明其促进成牙本质分化的能力要明显强于对照和VN。尽管2种蛋白均表现出优于对照组的矿化结节形成，但iMatrix-511展现出更强的钙化沉积效果。

结论：iMatrix-511在体外可以促进增殖并获得成牙本质分化的细胞表型，表明这种全新的材料可以作为牙本质再生的潜在候选者。
