这项研究利用妇女健康倡议(Women's Health Initiative,由美国国家心脏、肺和血液研究所资助)提供的8,155名黑人女性、3,494名西班牙裔女性和3,697名白人女性的遗传学样本和健康记录。在将CND和T2D女性与健康女性进行对比分析时,本文第一作者、人口健康和临床流行病学研究中心博士后研究人员Kei Hang K. Chan及研究小组发现,调节细胞粘附(细胞如何粘附在组织内)、钙信号(细胞如何沟通)、轴突导向(神经元如何找到与靶位点连接的途径)、细胞外基质(组织内的结构支持)和各种形式心肌症(心脏肌肉问题)的8种通路,存在关键的差异。

这些都是种族间普遍存在的。此外,研究小组发现,一些通路在T2D和CVD之间是种族特异性的。

Chan利用5种不同的方法,来对这些通路进行分析,只报道那些通过至少两种方法表现为重要显著性的通路。

从那里,通过对基因及其通路进行网络分析,进一步确定了可能是疾病“关键驱动因子”的基因。本文列出了它们中的“前十名”。

作者写道:“这些关键的驱动基因代表着关键的网络基因,当这些基因被扰乱时,可能潜在地影响参与CVD和T2D途径的大量基因,从而对疾病产生更强的影响。”

潜在的治疗靶点

为了评估这些基因是否可以作为关键的驱动因子,研究小组查阅了研究人员编写的有关“这些基因在健康小鼠模型中的重要性”的多个数据库。

在这篇论文中,依据它们与疾病如何合理相关,研究人员讨论了所涉及的通路。例如,轴突导向,众所周知参与发育中的胎儿如何建立神经系统,也涉及到维持胰腺中β细胞的机制发生,这是糖尿病的核心所在。这一通路如果发生分解,可以使细胞更容易受到攻击,影响糖的加工过程。

Liu说,随着这些通路和关键驱动基因的发现,现在有充分的机会来进行后续研究,从而使我们对于“这些通路在血管健康预后中的作用”,有更深入的理解,并设计和检验可能修复它们的治疗方法。

研究人员写道:“利用一种系统生物学框架,将人类和小鼠种群的GWAS、通路、基因表达、网络和表型信息进行整合,我们就能够深入了解内在的作用机制,并确定潜在的治疗靶点。”
