Heliyon：研究发现成釉蛋白C末端变体存在于人脂肪组织中

成釉蛋白（AMBN）启动子中的转录调节元件表明脂肪生成可能影响其表达。本研究旨在探究AMBN是否在脂肪组织中表达，并且在脂肪细胞分化过程中起作用。

通过实时PCR和ELISA检测脂肪组织和脂肪细胞中的AMBN表达。通过人皮下脂肪组织切片中的免疫荧光研究成釉蛋白的分布。通过[3H]-胸苷掺入测量类似于AMBN及其加工产物的重组蛋白对原代人前脂肪细胞和鼠3T3-L1细胞系增殖的影响。通过脂肪形成标志物PPARγ和瘦蛋白的表达谱以及脂质液滴的含量（油-红-O染色）评价对脂肪细胞分化的影响。

结果发现，AMBN在人脂肪组织，人原代脂肪细胞和3T3-L1细胞中表达。在人皮下脂肪组织中鉴定出AMBN蛋白的C末端和45bp较短的剪接变体。发现AMBN的表达在3T3-L1细胞分化期间增加4倍。重组AMBN的施用分别减少3T3-L1和人前脂肪细胞中的增殖，并增强PPARγ和瘦蛋白的表达。

总之，该研究在脂肪细胞中鉴定出AMBN C-末端变体，该变体可以由短剪接变体编码。在脂肪形成过程中AMBN的表达增加及其对脂肪形成因子的影响表明AMBN也在脂肪细胞发育中起作用。
