试验流程

这项研究是由邓迪大学研究小组与Wellcome Trust Sanger 研究所,以及北卡罗来纳大学协作完成。新研究结果发表在国际著名期刊eLIFE上。

细胞环境极其复杂:在任何一个时候,数千种不同的基因活化作为分子模板生成信使RNA (mRNA)分子,mRNA自身则是合成蛋白质的模板。然而并非所有的基因在所有细胞内总是处于活化状态。作为细胞分裂周期的一个组成部分当细胞生长和分裂之时,基因经常地开启和关闭。同样,在免疫系统和皮肤细胞中mRNA和蛋白质生成的模式也不相同。

该研究的领导者、邓迪大学生命科学学院Angus Lamond教授说:“我们能够超越以往详细地分析人类癌细胞中的蛋白质活性。由于无时无刻不在发生动态改变,如果我们想要了解癌细胞中的复杂过程,研究基因活性随时间的变化是非常重要的。”

“以往只能捕获平均时间上的这种活性快照,现在我们能够提供更加完整的图像。”


Lamond教授研究组首席研究员Tony Ly博士说:“这项工作增进了对mRNA水平与对应蛋白质产物复杂关系的理解。它也演示了如何能够检测到不同细胞类型和疾病状态之间,包括不同类型的癌症之间存在的细微但重要的差异。”

邓迪研究小组的这项研究工作提供了蛋白质水平上的新高分辨率基因表达图谱,也为未来开发更安全的新药带来了希望。几乎所有的药物都直接或是间接地影响着蛋白质。

对细胞蛋白质开展综合详细的分析,蛋白质组学正快速发展为超越基因组分析的下一个主要阶段,具有极大的潜力增进我们对于人类疾病的理解,帮助开发新治疗。
