Nature: 细胞自噬调控新老干细胞的代谢与功能

细胞自噬帮助造血干细胞清理线粒体的堆积,从而延长造血干细胞的功能抗衰老。

随着年龄增长,造血干细胞逐步失去造血机能,从而导致各类疾病。细胞自噬和健康及寿命密切相关,是保护造血干细胞免受代谢失衡的重要机制。在这一期的自然杂志中, 作者Theodore T. Ho 们发现了不能进行细胞自噬的造血干细胞会有线粒体的堆积,并且持续处于一种代谢激活的状态,从而通过非正常的DNA修饰加速髓系(myeloid)细胞的分化,最终影响造血干细胞的自我增值能力。



有趣的是,老实验鼠的大多数造血干细胞都有上述这样的代谢和功能的缺陷。然而还有三分之一的造血干细胞仍然保持着与年轻老鼠类似的较高的细胞自噬能力,较低的代谢速度,以及正常的自我增值能力。他们的文章系统的证明了细胞自噬通过维持正常的线粒体的状态来抑制造血干细胞的代谢速度,从而保证造血干细胞随着年龄的增长依旧能够正常的自我增值对抗衰老。
