fMRI软件统计方法有误,4万篇文献可能被中招!

功能性磁共振成像(fMRI,functional magnetic resonance imaging)是一种神经影像学方式,其原理是利用磁振造影来测量神经元活动所引发之血液动力的改变。用于这种方法的非侵入性、没有辐射暴露问题等优点,被广泛应用于神经科学研究。然而近期来自麻省总医院的一组研究人员发现,fMRI数据处理最常用的软件包产生的假阳性率高达 70%,这令其所涉及的40,000项研究

功能性磁共振成像(fMRI,functional magnetic resonance imaging)是一种常用的功能神经影像学方式,其原理是利用磁振造影来测量神经元活动所引发之血液动力的改变。用于这种方法的非侵入性、没有辐射暴露问题等优点,被广泛应用于神经科学研究。然而近期来自瑞典和英国的一组研究人员发现,fMRI数据处理最常用的软件包(统计处理错误,应该采用非参检验的,采用了参数检验)产生的假阳性率高达 70%,这令其所涉及的40,000项研究受到质疑这一研究成果公布在6月28日的《美国国家科学院院刊》(PNAS)杂志上。

梅斯医学编辑在pubmed上检索发现,以"functional magnetic resonance imaging" or fMRI[tiab]进行检索,有39169篇文献涉及fMRI技术。

虽然,fMRI假阳性为大家所知,但是从来没有人怀疑技术本身。2010年,PNAS上发文也质疑假阳性太高的问题(PNAS:fMRI可探测记忆,受假阳性困扰)。

“虽然功能磁共振成像fMRI已经25岁了,但令人惊讶的是这种技术的最常见统计方法未经过真实数据的验证,”文章作者,瑞典林雪平大学的Anders Eklund表示。

功能性磁共振成像是近年来神经影像学研究的重大进展之一,已从实验室研究进入到临床实验阶段,对神经系统进行了详细的研究,在一些疾病的研究方面具有重要的作用。曾经科学家利用fMRI研究了人类思考某一具体的名词时大脑在哪个区域产生相对应的活动,预测人脑对具体字眼反应的大脑活动模式,揭示外在的事物如何转变为人脑的内在感受,十分有趣。

这种成像方法能将大脑划分为许多个小单元:voxels,通过这种方法来检测大脑活性。然后再利用软件来对这些小单元分类,寻找相似活性的“区域”。

Eklund和他的同事分析了来自500例健康对照的静息态(resting-state,就是大脑非任务状态)fMRI数据,然后随机分配到对照组和“实验”组,这些数据会被数千次分配到三个常见的软件包:PM、 FSL 和 AFNI中的一个,造成假阳性。为了减少假阳性,往往在研究时,还会引入任务态,增强对照,但即使如此,往往仍然有大量的脑区呈现相似的阳性信号,这些往往被研究人员认为“其它干扰”而去除。但是统计学本身带来的错误,一直被忽视。

除了高得惊人的假阳性率,研究人员还在AFNI 软件包中发现了一个bug:当他们利用软件的一个调试版本时,能减少10%的假阳性率。研究人员指出,大多数采用这种错误软件的研究数据不可用,因此数据不容易重新分析。

除此之外,最近的一项研究指出最强的血管反应匹配附近最强的神经活动,表明fMRI可告诉我们关于大脑某区域总体功能的许多信息。坏消息则是,fMRI永远无法精确绘制神经回路。

这组研究人员发现在感官刺激过程中,并未精确“调整”血流量增加以响应局部的神经活动,由此挑战了血管和局部神经反应紧密联系这一长期以来持有的观点。
