单羧酸转运蛋白4（MCT4）是癌细胞糖酵解代谢的一个关键部件，对癌细胞恶性生物学行为也同样重要。近日，广州中山大学附属肿瘤医院和山东大学附属省立医院的外科学者对MCT4在肝细胞癌（HCC）患者中的表达情况、预后价值及其在肝癌细胞增殖和转移中的作用进行了研究，探讨其与癌症发生分子机制的关系，相关研究成果在线发表于12月2日的Journal of Cancer Research and Clinica

单羧酸转运蛋白4（MCT4）是癌细胞糖酵解代谢的一个关键部件，对癌细胞恶性生物学行为也同样重要。

近日，广州中山大学附属肿瘤医院和山东大学附属省立医院的外科学者对MCT4在肝细胞癌（HCC）患者中的表达情况、预后价值及其在肝癌细胞增殖和转移中的作用进行了研究，探讨其与癌症发生分子机制的关系，相关研究成果在线发表于12月2日的Journal of Cancer Research and Clinical Oncology杂志上。

该研究对318例肝细胞癌患者行肝切除术的病理标本进行免疫组化染色来检测MCT4的表达情况，并通过肝癌细胞株培养对MCT4表达和细胞增殖、转移、侵袭之间的关系进行了验证。

研究结果显示，表达水平增高的MCT4参考性指示甲胎蛋白水平升高和更大的肿瘤大小，将导致更差的无病生存率(DFS)和总体生存率(OS)。多元回归分析显示MCT4表达水平是肝细胞癌患者无病生存率和总体生存率的一个独立预后因子。在91例接受肝动脉灌注化疗栓塞术（TACE）治疗后复发的肝细胞癌患者，降低的MCT4表达水平预测一种有效的治疗反应和改善的总体生存率。此外，在MCT4 siRNA细胞系实验中，癌细胞增殖、转移和侵袭与MCT4表达密切相关。研究还发现，基因敲除MCT4与下调的AKT磷酸化和低氧诱导因子HIF-1α有关。

这项研究认为，MCT4表达水平可预测肝细胞癌患者生存率和肝动脉灌注化疗栓塞术的治疗反应。此外，MCT4在肝癌细胞增殖、转移和侵袭中扮演重要角色。抑制MCT4表达可诱导HIF-1α失活及抑制AKT磷酸化。MCT可能是肝细胞癌一个潜在的药物作用靶点。
