J Dent Res:CPNE7可诱导牙本质过敏模型的生物性牙本质封闭

由于多种原因导致牙本质小管开放进而引起牙本质过敏时常发生。作者之前的研究发现,copine 7 (CPNE7)能够体内诱导间接盖髓模型牙本质的形成。这篇研究的目的是通过暴露的牙本质在不添加任何条件的情况下应用CPNE7,评估第三期牙本质形成的情况,也检测其是否会影响牙齿的微渗漏以及体外评估CPNE7分子在牙本质小管内的渗透性。

研究对5只比格犬的6个上颌切牙颈环区域制备类似于V类洞型,1侧的3个上颌切牙施以重组CPNE7蛋白刺激5 min作为实验组。另1侧视为对照组,预备后未施加EDTA和CPNE7刺激。缺损处未施以任何修复,并且所有比格犬在4周后处死。通过微渗漏测试设备和激光共聚焦扫描显微镜检测暴露牙本质区域的流体渗透性。通过扫面电子显微镜对第三期牙本质形成情况进行组织学分析和观察。

结果显示,由于合面牙本质小管或第一或第二期牙本质的不连续,导致第三期牙本质形成减弱了牙本质小管内流体的流动。比格犬前牙的体内牙本质敏感模型组织学结果显示,与对照组相比,施以重组CPNE7蛋白的牙齿在牙髓-牙本质边界处新形成了第三期牙本质。扫面电子显微镜分析表明,合面部位的牙本质小管内存在矿物质沉积。在渗透性测试结果中,施以重组CPNE7蛋白组的平均微渗漏值明显低于对照组(P < 0.05)。激光共聚焦扫描显微镜结果验证了结合rhodamine B的CPNE7的小管渗透性。

结论:CPNE7通过暴露较浅的牙本质小管诱导第三期牙本质的形成,进而降低牙本质的渗透性。
