研究进行了免疫荧光，免疫印迹，PAD活性测定和双向电泳，并开发了器官型培养模型。

结果：PAD1在腭中表达最高，而PAD2，PAD3和PAD4表达在皮肤中最高，表明PAD同工酶的组织特异性表达导致钙依赖性的差异。免疫印迹显示FLG单体及其降解产物和前体（proFLG）在皮肤中表达最丰富，但在腭中低表达，而在颊粘膜中仅检测到微弱的proFLG表达。 FLG和K1与PAD1共定位，并且可能在皮肤的角质化层中被瓜氨酸化；在腭表面未检测到这种共定位，并且在腭的颗粒中发现可能是瓜氨酸化和PAD1的pFL1点状存在。源自大鼠腭的器官型模型显示，PAD抑制降低了FLG的分解，增加了其与K1的结合以及上皮致密性，并且在染料渗透性测定中发现其降低了渗透性。相反，PAD刺激具有相反的效果。

瓜氨酸化可能是蛋白质修饰，其以与皮肤明显不同的方式，在维持口腔角质化粘膜的结构和功能中起重要作用。
