上皮细胞形成排列在器官外表面的组织。间充质细胞是通常参与伤口修复等过程的迁移性细胞。上皮-间质转化(epithelial-mesenchymal transition, EMT)是一个将上皮细胞转化成迁移性的间充质细胞的过程。癌症劫持了这一过程,并通过让它停止在中间阶段而形成转移性的杂合细胞而变得最具侵袭性。

莱斯大学的研究人员之前已针对癌症干细胞如何通过控制参与胞外信号转导、伤口愈合和胚胎发育的Notch信号通路而在彼此之间和与外部世界之间进行通信提出了多种理论,比如EMT理论、干性理论、癌症代谢理论和Notch-Delta理论。在之前的研究中,他们已发现激活这种信号通路的配体是如何发生差错从而让迁移性的癌细胞保持在一起并帮助它们附着到身体新部位的结缔组织上。

在真实的系统中,所有这些理论都存在关联。在这项新的研究中,这些研究人员证实在遭受应激时,JAG1蛋白参与所有的这些理论。此外,他们还首次描述了炎性蛋白(包括细胞因子IL-6)如何扩大Notch和JAG1蛋白之间的信号,从而有助于促进杂合的上皮-间充质癌症干细胞的形成。

这项新的研究将所有这些来之不易的理论结合在一起,从而针对肿瘤如何进展形成了一个连贯性的全貌:JAG1是头号嫌疑犯,炎性蛋白起辅助作用。

这些研究人员使用了一种新的炎性乳腺癌细胞的肿瘤类器官模型,其中这种模型富含肿瘤干细胞,并发现减少JAG1的存在会影响它们的癌症干细胞样特性和增殖能力。这项研究可能导致人们开发出追踪和破坏这些细胞的疗法。

具有间充质细胞特征的癌症干细胞倾向于停留在肿瘤的边缘,在那里,它们接触周围环境,特别是炎性蛋白。肿瘤内部的癌症干细胞往往会向外扩散,形成药物更难摧毁的杂合干细胞。

这也突显了JAG1作为一种调节分子起着至关重要的作用,它将肿瘤进展的所有不同方面连接在一起。

最后,JAG1也可能作为开发新的诊断方法的靶标。
