细胞和机体稳态的维持决定了衰老的进程。在细胞水平,机体平衡部分是通过细胞自噬来维持的。细胞通过该机制能够完成错误折叠蛋白的清除,细胞器的更新,以及降解成分的循环利用。随着细胞的衰老,细胞自噬的能力会逐渐下降。蛋白质和细胞器质量控制的失调是继发于自噬减少的衰老标志。然而,至今为止,在生物体中,自噬的生理以及分子调控机制仍旧没有完全研究清楚。 来自凯斯西储大学的研究人员发现,Kruppel样家族的转录因子对于线虫的自噬和健康寿命的调节起着重要的作用,其也能够调节哺乳动物衰老相关的血管老化。在不同的长寿线虫模型中,Kruppel样家族转录因子的缺失会导致自噬的缺陷以及寿命的缩短。相反,Kruppel样家族转录因子的过表达使线虫寿命以自噬依赖的方式有所延长。此外,研究人员发现,哺乳动物血管因子Kruppel样转录因子4能够保守地在老年小鼠中增强自噬,改善血管功能。在人类血管内皮细胞中,Kruppel样家族转录因子4的表达也随着年龄的增加而降低。 因此,Kruppel样转录因子家族构成调节自噬和长寿的转录调节点,其在小鼠血管系统中具有保守作用,并且对哺乳动物血管衰老有潜在的影响。 原始出处: P
