CLIN CHEM:基于纸基的血浆和循环肿瘤细胞微小RNA表达谱

常规分离的循环肿瘤细胞受白细胞和红细胞污染使得循环肿瘤细胞特异性分子特征的鉴定极为困难。一种纸基的提取方法可以从有限的生物样品中最大限度地回收miRNA并进行下游分子分析。

近日,临床化学杂志(Clinical Chemistry)在线发表了一篇关于关于循环肿瘤细胞检测的一项研究,报道了利用基于纸基的miRNA方法对血浆和循环肿瘤细胞进行检测的研究成果。

循环肿瘤细胞(CTC)的分子特征在监测转移性进展和描述转移性疾病特征的方面具有广阔的前景。然而,常规分离的循环肿瘤细胞受白细胞和红细胞污染使得循环肿瘤细胞特异性分子特征的鉴定极为困难。

研究人员采用使用纸基介质从微量的生物样品中,如从癌症患者血液中收集的的罕见循环肿瘤细胞中,对微小RNAmiRNA)进行有效提取。具体来说,工作流程包括使用Flinders Technology Associates (FTA)® 洗提卡的靠数字PCR激发的“分隔”方法从血浆和循环肿瘤细胞中提取和纯化miRNA

研究表明这种方法的敏感性可以检测到少至掺入人类血液的3个癌细胞的miRNA表达。使用这种方法,可以从从污染血液细胞排除背景miRNA表达,并且可以检测到源自乳腺癌和结肠直肠癌患者循环肿瘤细胞的特异性miRNA表达谱。从循环肿瘤细胞纯化期间分离出的血浆同样可以使用类似的基于纸基的方法处理而用于miRNA的检测,从而可以从单次抽血最大化地获得患者特异性的信息。

总体而言,这种基于纸基的提取方法是一个高效,成本效益较好的工作流程,可以从有限的生物样品中最大限度地回收miRNA并进行下游分子分析。
