已经证实自然老化损害骨骼肌的功能。我们也知道肌肉干细胞的数量和活动随着年龄的增长而衰退。然而,原因尚不完全清楚。在一项新的研究中,Karolinska研究所的研究人员已经研究了肌肉干细胞中积累的突变数量。

最令人惊讶的是大量的突变。我们已经看到一个健康的70岁的人如何在肌肉中的每个干细胞中积累了超过1000个突变,并且这些突变不是随机的,但是有一些区域是受到更好的保护的。

突变发生在自然细胞分裂期间,受到保护的区域是那些对细胞的功能或生存很重要的区域。尽管如此,研究人员还是能够发现这种保护随着年龄的增长而下降。

可以证明,这种保护减少了您变老的细胞,表明细胞修复其DNA的能力受损,而这正是我们应该能够用新药物影响的事物。

研究人员从完成这项研究的新方法中获益。这项研究是用培养的单干细胞进行的,为整个基因组测序提供足够的DNA。在骨骼肌组织中做到了这一点,这是绝对独一无二的,还发现基因突变的重叠非常少,尽管细胞之间的距离很近,这代表着极其复杂的突变负担。
