J Periodontol：振动增强hpdl细胞中PGE 2、IL-6和IL-8的表达

尽管与正畸力相结合的振动可以加速正畸牙齿移动，但是关于调节相关细胞反应的机制知之甚少。本研究的目的是研究机械振动与压力相结合是否能通过COX途径调节人牙周组织中环氧合酶（COX）-2 /前列腺素E2（PGE2）和白细胞介素（IL）-6以及IL-8信使RNA（mRNA）和蛋白表达。

尽管与正畸力相结合的振动可以加速正畸牙齿移动，但是关于调节相关细胞反应的机制知之甚少。本研究的目的是研究机械振动与压力相结合是否能通过COX途径调节人牙周组织中环氧合酶（COX）-2 /前列腺素E2（PGE2）和白细胞介素（IL）-6以及IL-8信使RNA（mRNA）和蛋白表达。

方法：在24，48和72小时时，将初培养的hPDL细胞暴露于机械振动，压缩力或二者的组合。使用定量实时聚合酶链反应（qPCR）测定COX-2，IL-6，IL-8，核因子κB配体的受体激活剂（RANKL）和骨保护蛋白（OPG）mRNA表达。通过酶联免疫吸附测定（ELISA）定量测试PGE2，IL-6和IL-8蛋白表达。为了证明PGE2，IL-6和IL-8的表达是否在COX依赖性途径中，用吲哚美辛处理hPDL细胞。为了确定PGE2是否刺激hPDL细胞表达IL-6和IL-8，将外源PGE2加入培养基中。

结果：机械振动和压缩力的组合协同上调RANKL / OPG，COX-2 / PGE2，IL-6和IL-8mRNA以及蛋白质表达。吲哚美辛显着减弱了用压缩力或机械振动结合压缩力刺激的细胞中PGE2，IL-6和IL-8表达的增加。此外，外源性PGE2增加hPDL细胞中IL-6和IL-8 mRNA和蛋白的表达。

通过环加氧酶途径的机制，机械振动可以在正畸牙齿移动期间增强压缩侧的牙槽骨吸收。
