首先建立了HS大鼠模型。使大鼠的平均动脉压降低至30mmHg 维持90分钟,然后复苏。大鼠在复苏时用青蒿琥酯(2.4或4.8mg / kg i.v.)或载体随机处理。 4小时后,评估器官​​损伤和功能障碍的参数。

此项研究结果显示:青蒿琥酯可以减轻由HS引起的多器官损伤和功能障碍。 RNA测序分析结果显示青蒿琥酯对Akt-存活途径的作用,导致白介素-1受体相关激酶1的下调。

使用蛋白质印迹分析,研究者们证实予以青蒿琥酯治疗的HS大鼠的增强了蛋白激酶B(Akt)的磷酸化(激活)和内皮一氧化氮合酶以及糖原合酶激酶-3β(GSK-3β)磷酸化(抑制)。

此外,青蒿琥酯减弱HS诱导的核因子κB的活化并减少促炎蛋白(诱导型一氧化氮合酶,肿瘤坏死因子-α和白介素6)的表达。

此项研究结果表明:青蒿琥酯通过涉及Akt-内皮一氧化氮合酶存活途径的活化以及糖原合酶激酶-3β和核因子κB的抑制的机制减弱与HS相关的器官损伤/功能障碍。尚需II期临床试验评价青蒿琥酯在创伤和严重出血的患者 的效果。
