Basic-helix-loop-helix (bHLH)转录因子在多种器官发育中起到了重要作用；然而，从没有关于牙齿特异性bHLH的报道。在这篇研究中，作者采用酵母双杂交系统通过筛选一个牙胚互补DNA（cDNA）文库检测出一个全新的牙齿特异性bHLH转录因子，作者将其命名为AmeloD。AmeloD可以与小鼠染色体1q32相匹配。系统发育分析显示，AmeloD属于achaete-scute complex-like ( ASCL)基因家族，并且是ASCL5的同系物。AmeloD特异性的表达于内釉上皮细胞(IEE)，但是它的表达在IEE细胞分化为成釉细胞后受到抑制。并且，AmeloD的表达与牙上皮细胞特异性细胞间黏附分子E-cadherin的表达模式相反。在牙上皮细胞系CLDE中过表达AmeloD会导致E-cadherin的表达抑制。研究发现，AmeloD可以与E-cadherin基因启动子近端区域的E-box cis调控元件相结合。这些结果表明，AmeloD可作为IEE细胞内E-cadherin转录的抑制剂。结论：AmeloD是一个全新的牙齿特异性bHLH转录因子，它可能通过抑制IE
