PNAS:CCR7 调节记忆CD8 T细胞稳态增殖的机制

CD8记忆T细胞的群体是异质性的,不同的亚群在寿命,功能以及分布上都有明显的差异。这些亚群能够通过CD27、CD28、CD62L、CCR7、CD103等表达量的差异进行区分。目前了解较多的是中央记忆T细胞(Tcm)以及效应性记忆T细胞(Tem)。一般来讲,Tcm细胞大量表达CD62L/CCR7/CD27/CD28,它们较多地分布于脾脏与淋巴结中,而且能够分泌较多的IL-2。Tcm的主要作用是在二次感染的时候能够迅速地增殖,而在稳态条件下则维持一定水平的自我更新。相比之下,Tem细胞中淋巴结的归巢因子与共刺激因子的表达量较低,而大量表达毒性蛋白。因此,Tem细胞主要存在于血液与非淋巴组织中。对于特定的二次感染,免疫应答依赖于Tcm与Tem的协同作用。

除此之外,在皮肤,大脑,肺部,胸腺,粘膜层以及肠道中存在着一定量的组织特异性记忆T细胞(Trm),它们能够保护局部组织的二次感染。理解这些不同亚群的CD8记忆 T细胞对与鉴定有效的短期或长期免疫保护机制具有重要的意义。

记忆T细胞的产生与维持依赖于IL-7与IL-15。IL-7对于记忆T细胞的存活以及从非造血干细胞来源分化过程十分关键。在脾脏或淋巴结中,IL-7主要由T区的成纤维网状细胞(FRC)分泌,这部分细胞同样分泌CCL19与CCL21,用于招募CCR7阳性的T细胞。因此,CCR7对于促进T细胞接受IL-7的刺激十分关键。

为了进一步理解CCR7对于维持记忆T细胞的存活以及分化的作用,来自耶鲁大学的Susan M. Kaech课题组进行了深入研究,相关结果发表在最近一期的《PNAS》杂志上。

首先,作者猜想记忆T细胞的寿命与其归巢进入脾脏的T区并与IL-7基质细胞的接触具有一定的相关性。为了验证这一猜想,作者将野生型小鼠或CCR7缺失突变体小鼠的CD8 T细胞转入受体小鼠中,之后进行LCMV的感染。之后,作者分别于8天以及35天之后取出小鼠的脾脏并进行组化染色分析。结果显示,CCR7缺失突变的T细胞进入脾脏T区的能力受到了明显的影响。

之后,作者利用类似的方法进行刺激,并分析了小鼠脾脏、淋巴结以及各组织器官中供体CD8 T细胞的表型特征,发现CCR7的缺失并不会影响CD8 T细胞的扩增,但CD8 T细胞向记忆性T细胞的分化与存活会明显升高。这一差异部分地受到Bim低表达的影响。进一步,作者发现CCR7缺失之后KLRG1loIL-7Rhi的CD8记忆性前体T细胞的存活以及其向记忆T细胞的分化也会相应增强。此外,CCR7缺失突变体细胞相比野生型细胞的稳态增殖速率也明显更高。

综上,作者发现CCR7信号对于CD8记忆T细胞的分化、存活以及增殖都有一定的负向调控作用。
