在骨组织工程过程中,生物材料的骨免疫调节性能对干细胞的成骨分化非常重要,它决定了骨再生的结果。镁(Mg)是一种可生物降解、生物相容性好的金属,具有骨传导性,一直被认为是一种很有前途的骨生物材料。然而,Mg的高降解率会导致过度炎症,从而限制了其在骨组织工程中的应用。重要的是,已经利用不同的涂层或镁合金来降低降解率。

 

之前的一项研究证明,Mg支架的β-TCP涂层可以调节Mg基生物材料的骨免疫调节性能,为成骨创造有利的免疫微环境。然而,Mg离子本身的骨免疫调节特性尚未得到探讨。本研究系统地研究了Mg离子在巨噬细胞和骨髓干细胞(BMSCs)参与下的骨免疫调节特性,研究结果已在线发表于Biomed Res Int。

结果发现,微尺度Mg离子(100mg/L)具有有利于骨形成的骨免疫调节特性。具体来说,微尺度Mg离子通过抑制TLR-NF-κB信号通路,诱导巨噬细胞的M2表型变化和抗炎细胞因子的释放。微尺度的Mg离子还刺激了巨噬细胞中骨诱导分子的表达,而Mg离子/巨噬细胞条件培养基通过BMP/SMAD信号通路促进了BMSCs的成骨。

综上所述,这些研究结果表明,控制Mg离子浓度可以赋予Mg生物材料有利的骨免疫调节性能,从而为改善和修饰Mg基骨生物材料的效果提供了基本证据。
