J Cell Physiol:组蛋白脱甲基酶KDM7A是乳腺癌干细胞维持和细胞凋亡抑制所必需的

已知组蛋白脱甲基酶KDM7A可调节哺乳动物的神经元分化和发育。在这项研究中,我们发现KDM7A也是乳腺癌干细胞(BCSCs)维持所必需的,研究结果已在线发表于J Cell Physiol。

已知组蛋白脱甲基酶KDM7A可调节哺乳动物的神经元分化和发育。在这项研究中,我们发现KDM7A也是乳腺癌干细胞(BCSCs)维持所必需的,研究结果已在线发表于J Cell Physiol。

沉默KDM7A可显著降低BCSCs数量和体外乳腺球的形成,并抑制体内乳腺肿瘤的生长。恢复KDM7A的表达可以挽救干细胞维持的缺陷。机制分析表明,KDM7A上调了干细胞相关因子KLF4和c-MYC以维持BCSCs。此外,KDM7A的敲除通过降低乳腺癌(BrCa)中BCL2的表达和BAD的磷酸化来促进细胞凋亡。此外,恢复KDM7A和BCL2的表达可解救乳腺癌的凋亡抑制,说明KDM7A通过上调乳腺癌中BCL2的水平抑制了乳腺癌的凋亡。

 

综上所述,该研究结果表明,KDM7A能促进乳腺癌中癌症干细胞的维持和凋亡抑制。
