科学家们花了几十年时间研究肿瘤细胞的性质,但很少有人去观察肿瘤周围那些正常的组织发生了什么。事实上,健康组织中的细胞同样以惊人的频率发生突变。这些突变中许多都与异常的细胞行为,比如分化缺陷和过度增生有关,但是大多数正常组织并不会因此出现明显的病理表型。那么,这些组织如何在不断的突变中保持正常?近日,来自耶鲁大学的研究人员在皮肤细胞中发现,癌细胞附近未受影响的正常细胞并不会坐以待毙,而是像细胞警察一样,积极纠正邻近癌细胞所造成的组织缺陷。这项研究结果发表在8月2日的Nature期刊上。

该研究由耶鲁大学医学院的Valentina Greco和福瑞德哈金森肿瘤研究中心的Slobodan Beronja所领导。研究生Samara Brown和Cristiana Pineda是该研究的共同第一作者。Samara Brown说:“正常细胞甚至能够将突变细胞从组织中抓出来,并清除突变细胞留下的垃圾,以保持组织的健康和功能。”

Pineda补充说:“我们发现了一个动态的,积极的校正过程,在不同的突变系统中,甚至是突变无关的模型中存在。”

耶鲁大学研究小组观察了小鼠组织中损伤的细胞或激活的癌基因,并使用新型活体成像技术来研究周围细胞的行为。他们发现这些野生型细胞对于消除肿瘤以及校正损伤细胞引起的异常是必需的,在这些细胞不存在的情况下肿瘤发生明显进展。研究揭示了皮肤上皮细胞在面临突变和非突变的异常状况时所具有的出人意料的适应性,并阐明了恢复组织稳态的动态细胞行为。
