研究从6名健康个体口内提取混合唾液过夜制成生物被膜,以此来反应生物被膜成熟的早期阶段。首先,研究者们检测了生物被膜细胞的精氨酸摄入以及所产生的代谢产物。随后,向生物被膜预先添加75 mM精氨酸,在0, 6, 20和48 h后再添加15 mM的蔗糖,检验精氨酸对pH动力学的影响。另外,在每个时间节点检测pH值,并且收集生物被膜进行16S序列分析和靶向精氨酸的定量分析,上清液用来进行蛋白组学研究。

结果显示,在生物被膜中仅添加蔗糖导致pH值从7持续降至4.5,而预先添加精氨酸,在6, 20, 和48 h后再施加蔗糖刺激则使pH值恢复较高水平。在生物被膜细胞中检测到精氨酸,表明其可以主动吸收;在上清液中检测到精氨酸分解代谢产物瓜氨酸,鸟氨酸和腐胺,表明其可以活性代谢。序列分析揭示了在添加精氨酸的生物被膜微生物群落结构的转变以及种属多样性的增加。

结论:研究表明,精氨酸通过重塑口腔微生物群落可以提高pH内稳定。
